PARP 抑制剂在妇科肿瘤治疗中的研究进展

摘要(Abstract):

PARP抑制剂(PARPi)作为聚焦DNA损伤应答(DDR)通路的范例,依照“合成致死”这一遗传学原理,彻底扭转了特定类型癌症的治疗格局,在有同源重组修复(HRR)缺陷的肿瘤细胞中,PARPi通过抑制PARP酶活性及“PARP捕获”手段,造成DNA单链断裂,选择性诱发肿瘤细胞死亡。自首个PARPi奥拉帕利(Olaparib)获批起,这类药物在卵巢癌、乳腺癌等癌症治疗上取得了显著成效。本文系统回顾PARPi的作用机制、在主要癌种中的临床应用进展、生物标志物策略、联合治疗探索,对未来发展进行展望,旨在为这个领域的临床实践和未来研究提供参考。
